Pure water distribution system in pharmaceutical company

Dear all

I need information about purified water loop in pharmaceutical company.  PW water with USP (United State Pharmacopeia) specs (i.e., EC<1.3) is produced in a reverse osmosis package according to GMP (Good manufacturing practice) guidelines. Then, the product purified water is circulated continuously in a distribution loop that consists of a storage tank, pump, piping, distributor equipment and required equipment for maintaining water quality.

I need to know more about the mentioned distribution loop. What are the best design criteria for this loop? I need information specially about maintaining water quality. Can CO2 absorption change the water quality? Any information and document can be helpful for me.

However, how can we prevent CO2 absorption in PW water when the water temperature is about 25 oC? Besides, I think CO2 absorption is more feasible when ozone is injected in water. Because product gas of ozone generator contains CO2 gas.
However, would you please introduce me any reference that described all of the mentioned criteria in detail?

Surely CO2 will change water characteristic. No free CO2 allowed in EDI as it will change FCE(feed conductivity equivalence). As stated above by others velocity (return water) is important. You will have to ensure zero dead lag and bacteria free condition.

pure water distribution system is very impotant part in pharmaceutical industries. water distribution system prevent micro vial contamination in purified water. pipe line material , uv system, velocity of water. pressure of water and temperature of water and dead legs are important factors for any guidelines.

You have Water conductivity as per USP guidelines,
Other required parameters are as follows:
pH 5.0 – 7.0
Conductivity ≤ 1.3 @ 25ºC µS/cm
TOC < 500 Ppb
Microbial Count < 100 cfu/ ml.

CO2 presence will make water more acidic, should be maintained at pretreatment scheme.

Dear Hossein,

of course Carbon Dioxide affects the quality of water, due to the fact that the reaction between CO2 and water produce carbonic acid (H2CO3 which dissociate into H+ and HCO3-). This reaction increases significantly the electrical conductivity, and if you have a value < 1,3 µS, this means that the PW recirculates approximately at 25°C.

The guidelines suggest to keep the water at constant temperature (usually for PW is 16 - 18°C), the speed inside distribution pipe must be > 1 m/s, continuos control of electrical conductivity shall be installed on the loop and a UV lamp to control bacterial growth.

If you can provide further information, it could be possible to give you more hints about the system and its maintenance.

Carbonic acid will reduce the pH slightly. I do not think it would affect the quality of water used for formulation. Also before RO, we have to treat the water (disinfection etc), but higher concentration of any chemical disinfection will degrade the quality of RO membrane. But usually UV or O3 treatment will eliminate this. But suspended solids is a strict no.
